Are these direct plug and play gauges or do I need anything else, plugs or wiring to use on a 2007 Harley Davidson Road Glide ?
This kit will plug straight into the OEM wire harness on your bike. Assuming the instruments on your machine are completely stock, you need not purchase any additional parts to have it function as advertised.
I would think if these were waterproof that would have been advertised? Are these waterproof?

All Koso instruments possess an IP66 rating, which is more than enough for use on these machines. They can withstand 3 minutes of water sprayed at 100 liters per minute and 100 kPa at a distance of 3 meters through a 12.5mm nozzle. Basically, so long as you're not blasting them directly with a power washer from nearby, they won't ingest water.
Are the gauges only in red or are there other colors?

These gauges can be switched between red and blue lighting on the fly. That is to say, one kit allows you the options of either color at any time you want.
